OPTIMASI FORMULA SEDIAAN GEL EKSTRAK METANOL HERBA ASHITABA (Angelica keiskei) DENGAN METODE SIMPLEX LATTICE DESIGN
ABSTRAK
Sediaan gel antijerawat yang saat ini beredar memiliki sejumlah
masalah seperti resistensi dan memicu iritas kulit. Kandungan kalkon pada
ekstrak ashitaba dapat beraktivitas sebagai antibakteri penyebab jerawat.
Potensi antibakteri ekstrak ashitaba perlu dikembangkan menjadi suatu
sediaan, salah satunya sediaan gel. Tujuan penelitian ini adalah
menenetukan konsentrasi HPMC dan carbopol sebagai gelling agent untuk
mendapatkan formula optimum sediaan gel ekstrak metanol herba ashitaba
serta menentukan hasil evaluasi sifat fisiknya. Hasil deklorofilasi herba
ashitaba diekstraksi dengan metanol menggunakan metode sonikasi.
Selanjutnya ekstrak metanol herba ashitaba diuji pH, kadar air, dan
metabolit sekunder seperti alkaloid, flavonoid, tanin, saponin, dan
steroid/triterpenoid. Selanjutnya dioptimasi sediaan gel ekstrak metanol
herba ashitaba. Optimasi formula dilakukan dengan metode Simplex Lattice
Design dengan pH, daya sebar, daya lekat, dan viskositas sebagai parameter
respon. Hasil pengujian ekstrak metanol herba ashitaba diperoleh pH
sebesar 4,713?0,577, kadar air sebesar 1,24?47,085 %, dan positif
mengandung alkaloid, flavonoid, tanin, dan steroid. Hasil optimasi formula
sediaan gel diperoleh kombinasi HPMC dan carbopol masing-masing
sebesar 2,125% dan 0,875%. Sedangkan hasil evaluasi diperoleh nilai pH
sebesar 5,73?0,22; daya sebar sebesar 4,119?0,06 cm; daya lekat sebesar
44,67?3,09 detik; dan viskositas sebesar 5.935,33?575,20 cP. Evaluasi
akhir diperoleh bentuk gel yang kental, berwarna hijau-kecoklatan, dan
beroma khas, homogen, dan tidak terjadi sineresis. Berdasarkan hasil yang
diperoleh, sediaan gel ekstrak metanol herba ashitaba memiliki sifat fisik
yang baik.

ABSTRACT
Anti-acne gel preparations that are currently circulating have a number
of problems such as resistance and trigger skin irritation. The chalcone
content in ashitaba extract can function as an antibacterial that causes acne.
The antibacterial potential of Ashitaba extract needs to be developed into a
preparation, one of which is a gel preparation. The purpose of this study was
to determine the concentration of HPMC and carbopol as gelling agents to
obtain the optimum formula for ashitaba herb methanol extract gel
preparations and to determine the results of evaluating their physical
properties. The result of dechlorophyllation of ashitaba herb was extracted
with methanol using the sonication method. Furthermore, the methanol
extract of Ashitaba herb was tested for pH, water content, and secondary
metabolites such as alkaloids, flavonoids, tannins, saponins, and
steroids/triterpenoids. Furthermore, the preparation of ashitaba herb
methanol extract gel was optimized. The optimization formula was carried
out using the Simplex Lattice Design method with pH, spreadability,
adhesion, and viscosity as response parameters. The results of the ashitaba
herb methanol extract test obtained a pH of 4,713?0,577, a water content of
1,24?47,085%, and positively contained alkaloids, flavonoids, tannins and
steroids. The results of the optimization of the gel preparation formula
obtained a combination of HPMC and carbopol of 2,125% and 0,875%,
respectively. The evaluation results obtained a pH value of 5,73?0,22;
spreading power of 4,119?0,06 cm; adhesiveness of 44,67?3,09 seconds;
and a viscosity of 5,935,33?575,20 cP. The final evaluation was obtained in
the form of a thick gel, brownish green in colour, and had a distinctive
odour; homogeneous, and no syneresis occurs. Based on the results
obtained, the ashitaba herb methanol extract gel preparation had good
physical properties.
