UJI DIABETES MELITUS SECARA NON-INVASIF BERBASIS KADAR DAN PROFIL PROTEIN SALIVA

Latar belakang: Diabetes merupakan penyakit metabolik yang ditandai dengan keadaan hiperglikemia. Saat ini terdapat 40,9 juta penderita diabetes dan diprediksikan pada tahun 2025 akan meningkat menjadi 70 juta. Untuk menekan angka tersebut, diperlukan pola hidup sehat dan pemeriksaan rutin gula darah. Pemeriksaan glukosa darah dilakukan dengan proses pengambilan darah. Cara ini merupakan tindakan invasif yang memprovokasi nyeri, sehingga diperlukan prosedur non-invasif lainnya seperti pemeriksaan saliva.
Tujuan: Untuk mengetahui perbedaan kadar dan profil protein saliva pada individu sehat dan penderita diabetes.
Metode: Penelitian ini merupakan studi komparatif. Sampelnya adalah saliva yang berasal dari 20 orang dan terbagi menjadi 2 kelompok. Kelompok penderita diabetes (D), dan kelompok individu normal (N). Saliva yang terkumpul disentrifugasi dan diambil supernatannya, lalu dilakukan proses pengukuran kadar protein saliva menggunakan spectrophotometer NanoDrop. Profil protein saliva dianalisis menggunakan SDS-PAGE. Uji statistik kadar protein saliva antara individu sehat dan penderita diabetes dilakukan dengan uji Independent Sample T-test.
Hasil: Jumlah rata-rata kadar protein saliva pada kelompok penderita diabetes sebesar 2932±526μg/ml dan kelompok individu non-diabetes 2005±253μg/ml. Secara statistik peningkatan tersebut tidak signifikan (P>0,05). Pada profil protein saliva ditemukan 3 band peptida dengan berat molekul antara 25-35kDa pada penderita DM.
Simpulan: Terdapat kecenderungan adanya peningkatan kadar protein saliva pada kelompok DM. Namun, secara statistik peningkatan tersebut tidak signifikan. Pada profil protein ditemukan adanya perbedaan band peptida saliva antara penderita DM dan individu normal.

NON-INVASIVE DIABETES MELLITUS TEST BASED ON THE LEVEL AND PROFILE OF SALIVARY PROTEINS

Background: Diabetes is a metabolic disease characterized by hyperglycemia state. Currently there are 40.9 million diabetics in 2025 and is predicted to increase to 70 million. To suppress these figures, it takes a healthy lifestyle and regular checks of blood glucose. Blood glucose tests performed with using blood. It is an invasive procedure which provokes pain, so it requires a non-invasive procedures such as salivary examination.
Objective: To determine differences in the levels and profiles of salivary protein in healthy and diabetics individuals.
Method: This study was an comparative study. The sample was the saliva of 20 people that was divided into 2 groups. Groups of diabetics (D), and a group of normal individuals (N). The collected saliva was centrifuged and the supernatant was taken, and measurement process of the saliva protein levels was conducted using NanoDrop spectrophotometer. Salivary protein profiles were analyzed using SDS-PAGE. Statistical test between the healthy and diabetes idividuals saliva protein levels was performed by Independent Sample T-test.
Result: The average amount of saliva protein levels in group of patients with diabetes was 2932±526μg/ml and in group of non-diabetic individuals was 2005±253μg/ml. The increase is not statistically significant (P> 0.05). In the salivary protein profiles there was 3 peptide band with a molecular weight between 25-35kDa in diabetic individuals.
Conclusion: There is a tendency for increased levels of salivary proteins in the diabetic group. However, this increase was not statistically significant. There’s a differences of salivary peptide band between diabetic and normal individuals.
