PENGARUH EKSTRAK RAMUAN SYZYGIUM POLYANTHUM, IMPERATA CYLINDRICA, CANTELLA ASIATICA, ANDROGRAPHIS PANICULATA TERHADAP HISTOPATOLOGI GINJAL TIKUS (RATTUS NOVERGICUS) HIPERURISEMIA

PENGARUH EKSTRAK RAMUAN SYZYGIUM POLYANTHUM, IMPERATA CYLINDRICA, CANTELLA ASIATICA, ANDROGRAPHIS PANICULATA TERHADAP HISTOPATOLOGI GINJAL TIKUS (RATTUS NOVERGICUS) HIPERURISEMIA

Latar Belakang : Ginjal adalah organ yang berperan penting dalam pengaturan homeostasis, mengekskresikan senyawa asing atau toksik, dan sisa metabolisme tubuh seperti asam urat, urea, dan kreatinin dalam tubuh melalui urin. Ekstrak ramuan yang terdiri atas Syzygium polyanthum, Imperata cylindrica, Cantella asiatica, Andrographis paniculata digunakan untuk menurunkan kadar asam urat. Tujuan penelitian ini adalah mengetahui pengaruh pemberian ekstrak ramuan khas Lombok tersebut terhadap histopatologi ginjal pada tikus Rattus norvegicus setelah pemberian 14 hari.
Metode : Penelitian ini merupakan penelitian eksperimental laboratoris dengan desain post test only control group. Tikus yang digunakan berjumlah 24 ekor tikus jantan yang dibagi menjadi 4 kelompok yaitu kelompok kontrol negatif, kelompok kontrol positif (allopurinol 5 mg/200grBB), kelompok perlakuan dosis I (29 mg/200grBB) dan perlakuan dosis II (58 mg/200grBB). Semua kelompok diberi pakan diet tinggi kolesterol dan purin serta diinduksi propiltiourasil 10 mg/KgBB dan kalium oksonat 42 mg/200grBB. Tikus diterminasi pada hari ke-15 dan diambil sampel organ ginjal untuk dijadikan preparat. Preparat histopatologi diwarnai dengan pewarna HE dan diamati secara mikroskopis. Grading kerusakan ginjal yang terjadi menggunakan kriteria 0 = normal, 1 = lesi tanpa nekrosis, 2 = nekrosis. Data diolah menggunakan uji Fisher Exact.
Hasil : Hasil penelitian menunjukkan bahwa terdapat peradangan pada semua kelompok tikus dan kerusakan paling luas terdapat pada kelompok kontrol positif yaitu terjadi nekrosis. Hasil uji Fisher Exact didapatkan nilai p = 0,098 yang menunjukkan tidak ada perbedaan yang bermakna pada masing-masing kelompok penelitian.
Kesimpulan : Pemberian ekstrak ramuan Syzygium polyanthum, Imperata cylindrica, Cantella asiatica dan Andrographis paniculata tidak mempengaruhi kerusakan histopatologi ginjal tikus.

Effect of Syzygium polyanthum, Imperata cylindrica, Cantella Asiatica, Andrographis paniculata Extract on Renal Histopathology of Hyperuricemia Induced Rats (Rattus norvegicus)

Background: Kidney is an organ that plays an important role in regulating homeostasis, excrete the foreign or toxic substances, and metabolism product such as uric acid, urea, and creatinine in the body through the urine. A remedy consisting of Syzygium polyanthum, Imperata cylindrica, Cantella asiatica, Andrographis paniculata has been used in lowering uric acid levels. The aim of this study was to examine the effect of a traditional herb extract from Lombok on renal histopathology of Rattus norvegicus after 14 days of treatment.
Methods: This was a laboratory experimental research with post test only control group design. 24 male rats were divided into 4 groups: negative control group, positive control group (allopurinol 5 mg/200grBW), the first dose treatment group (29 mg/200grBW) and the second dose treatment group (58 mg/200grBW). All groups were fed by high cholesterol and purine diet and induced by propylthiouracil 10 mg/200grBW and potassium oxonate 42 mg/200grBW. The rats were terminated on day 15 and kidneys were taken to be used as preparations. Histopathology were prepared & stained by using HE stain and observed microscopically. Grading of kidney damage that occurs using criteria 0 = normal, 1 = lesions without necrosis, 2 = necrosis. The data were analyzed using Fisher Exact test.
Result: Histopathology inflamation occured in all group and the most severe necrosis found in positive control group. P value shown no significant difference among administration groups (p = 0,098).
Conclusions: Administration of Syzygium polyanthum, Imperata cylindrica, Cantella asiatica and Andrographis paniculata extract did not affect histopathology profile of Rattus norvegicus kidney.
